The dependence of the specific activity of urease upon the apparent absolute enzyme concentration.

چکیده
If the activity of an enzyme preparation is determined under conditions in which a further increase in substrate concentration is without demonstrable effect, all other factors being held constant, it is ordinarily assumed that the specific activity of the enzyme, expressed in terms of arbitrary units per unit weight of the enzyme, is independent of the absolute enzyme concentration (1, 2). However, with urease solutions stabilized with hydrogen sulfide or cysteine (1) we have observed that the specific activity of a given urease preparation, when determined under the above conditions, increases with decreasing apparent enzyme concentration over a wide range of concentrations and that this increase in specific activity proceeds with a measurable velocity at temperatures above 15”. This phenomenon was observed with crude urease preparations, such as jack bean meal, and with two, three, and seven times recrystallized urease. Since little or no difference was observed in the behavior of three and seven times recrystallized urease, the data presented in this paper are limited to those obtained with thrice recrystallized preparations. Urease activity was determined by a modification of the procedure described by Van Slyke and Cullen (3). The precision of the modified procedure was f2 to 3 per cent.

In Vivo, after administration of treatment, concentrations of antimicrobial agents will reduce to sub-inhibitory levels (sub-MIC) and may therefore affect the properties of target bacteria. The purpose of the present study was to investigate the in vitro effect of sub-minimum inhibitory concentrations (sub-MIC) of ampicillin, gentamicin and nalidixin acid on morphology, growth, ammonium produc...
متن کاملUrease Activity Protection With EDTA Against Nanoparticles (Fe2O3 and Fe3O4) Inactivation
In this study the effects of Fe2O3 and Fe3O4 magnetic nanoparticles and EDTA on urease activity was investigated. The effect of nano-Fe2O3 and nano-Fe3O4 on urease activity were investigated. Urease activity was studies by UV-Vis spectrophotometry at 40 °C at pH = 7.2 using sodium phosphate as buffer. Measurements were carried out using 0.075 mg/ml of urease and a range of nano-Fe2O3 and nano-F...

Urea is one of the most commonly used nitrogen fertilizers in agricultural lands. The fate of this fertilizer in soils is greatly related to the soil urease enzyme activity, while this enzyme commonly exists in the adsorbed state on the surface of soil colloids, and especially clay minerals. In order to examine the kinetic properties of free and adsorbed Urease on the surface of sepiolite and v...
متن کاملKinetic Investigation of Myeloperoxidase upon Interaction with Copper, Cadmium, and Lead Ions
Background: Myeloperoxidase (MPO), which is abundantly expressed in neutrophils, catalyzes the formation of a number of reactive oxidant species. However, evidence has emerged that MPO-derived oxidants contribute to tissue damage and initiation and propagation of inflammatory diseases, particularly, cardiovascular diseases. Therefore, studying the regulatory mechanisms of the enzyme activity is...
متن کاملUltrasound effects on the activity of free and immobilized urease on the magnetic nanoparticles and silica gel
In this study, the effect of low-frequency ultrasound on the structure and activity of urease enzyme has been investigated. For this purpose, the enzyme was exposed to ultrasound with 20 kHz frequency at different time intervals. The urease enzyme was immobilized on the surface of magnetic nanoparticles (Fe2O3), and activated silica gel with 89% and 66% efficiency respectively. The enzyme activ...
